Introduction: Determination of total proteins in cerebrospinal fluid (CSF) is valuable in the diagnosis of conditions such as bacterial and viral meningitis, encephalitis, tumours of the central nervous system (CNS)and cerebral hemorrhage.
Aim: The aim of this study was to compare the pyrogallol red-molybdate (PRM) method for determination of proteins in CSF with that of 3 % sulfosalicylic acid solution (SA).We also compared the results obtained with these methods in infectious diseases of the CNS.
Materials and methods: Total protein concentrations were measured in 45 CSF samples of patients hospitalized in the University Hospital for Infectious Diseases ''Dr. Fran Mihaljević''. The PRM assays were performed by the biochemical analyzer Olympus AU400 using both Olympus and Randox reagents, and the SA assay by an Iskra photometer. A set of solutions of known protein concentration (Kjeldahl) was used for determination of accuracy of the methods. The reproducibility was determined using a CSF sample.
Results: The results were statistically analyzed with linear regression analysis and the correlation coefficients were calculated. The correlation coefficients and the linear regression lines were: Olympus vs. Randox r =0,9924 , (y=0,9444x + 74,042); Randox vs. SA r=0,9778 (y=0,8333x - 65,372) and Olympus vs. SA r = 0,9749 (y= 07907x - 6,355). The imprecision (CV %) and inaccuracy (R%) for the assays were as follows : Olympus (1,70; 1,58), Randox (0,71; 0,02), SA (7,39; 3,72).
Conclusion: Linear regression analysis and correlation coefficients show a high correlation between all assays. Precision and accuracy were better with the PRM reagents (Olympus and Randox) than with SA.
